ATTO-conjugated antibodies   |   Guinea pig-raised polyclonals

Live cell flow cytometry with FITC-conjugated antibodies

Alomone Labs antibodies are mostly polyclonal and are raised in rabbit and guinea pig.

We choose the epitope for immunization very carefully, taking into account two main parameters:

✓ Immunogenicity
✓ Specificity of the epitope

Before immunization, we run a search (blast) to verify that the chosen epitope has maximum homology with other species and minimum homology between members of the same family, or other proteins.

All antibodies are affinity purified on immobilized antigens (the injected peptides). Antibodies are characterized and validated in-house by western blot analysis. All lots are tested and undergo strict quality control.

Reliability, reproducibility & results

The negative control antigen

The negative control antigen, is the peptide injected to immunize animals. We routinely use the negative control antigen to determine antibody specificity. We believe in the quality and reproducibility of our Antibodies. Therefore, for each polyclonal antibody, we also provide you with the negative control antigen (FREE of charge). We encourage you to use the negative control antigen in your experiments.

Live cell imaging with “extracellular” antibodies

Extracellular antibodies recognize proteins through extracellular domains. You can monitor cell movement, study protein transport across the cell, and investigate protein internalization.

✓ Work with live cells
✓ Cell permeabilization is unnecessary
✓ Cell fixation not required
✓ Cell-surface detection of protein

Innovative platforms

Antibodies developed for multiplex staining

Alomone Labs has developed two innovative platforms for studying protein-protein interaction and/or protein co-localization:

ATTO-conjugated antibodies

Primary antibodies conjugated to ATTO fluorescent dyes: These antibodies are developed for immunohistochemistry (IHC) and immunocytochemistry (ICC) applications.

✓ Direct immuno-fluorescence
✓ Secondary antibodies are not necessary
✓ Ideal for multiplex staining with same species/host antibodies

Multiplex staining of mGluR1 and TRPC3 in mouse cerebellum.Anti-mGluR1 (extracellular)-ATTO Fluor-488 Antibody (#AGC-006-AG)

Multiplex staining of CaV1.2 and GABA(A) α1 Receptor in rat cerebellum.Anti-GABA(A) α1 Receptor (extracellular)-ATTO Fluor-488 Antibody (#AGA-001-AG)

Guinea pig-raised polyclonal antibodies

The majority of commercially available primary antibodies are rabbit polyclonals or mouse monoclonals. Guinea pig-raised polyclonal antibodies offer greater flexibility in multi-immunostaining.

Live cell flow cytometry with FITC-conjugated antibodies

Antibodies conjugated to FITC dye are specifically developed for direct flow cytometry. They undergo testing with appropriate isotype controls. The FITC dye is conjugated to “extracellular” antibodies enabling live cell detection of proteins.

✓ Time saving
✓ Works with live cells
✓ Cell permeabilization is unnecessary
✓ Cell fixation not required
✓ Cell-surface detection of protein
