Current amplitudes were plotted as a function of time. Membrane potential was held at –80 mV and oocytes were stimulated by a 100 ms voltage ramp to 0 mV. 50 nM κ-Conotoxin-RIIIJ were perfused as indicated by the bar (green) at -15 mV for 160 sec. B. Scientific background
κ-conotoxin RIIIJ (κ-M-RIIIJ), is a peptide toxin belonging to the conotoxin M superfamily, and was originally isolated from Conus radiatus (Rayed cone) venom. This conopeptide inhibits the homotetrameric human KV1.2 channels (IC50 = 33 nM)3. κ-M-RIIIJ was found to exert cardioprotective effects, presumably due to the high affinity of this peptide for mammalian KV1.2 and KV1.2- containing heteromultimers3.
Voltage-gated K+ channels represent a structurally and functionally diverse group of membrane proteins. These channels establish the resting membrane potential and modulate the frequency and duration of action potentials in nerve and muscle1,2.
Target KV1.2 K+ channels
