- Alomone Labs ω-Conotoxin MVIID blocks CaV2.2 channels heterologously expressed in Xenopus oocytes.A. Time course of ω-Conotoxin MVIID (#STC-751) action on CaV2.2 maximum currents. Membrane potential was held at -100 mV and cells were stimulated by a 100 ms voltage ramp to +40 mV in the presence of 2 mM Ba2+. 0.2 µM (green) and 1 µM (red) ω-Conotoxin MVIID were perfused as indicated by the bar, each for 200s. Scientific background ω-conotoxins are small, disulfide-rich peptides from the venoms of predatory cone snails that inhibit neuronal voltage-gated Ca2+ channels involved in neurotransmitter release. ω-conotoxin-MVIID (ω-CTx-MVIID) was identified by sequence homology from Conus magus (Magician's cone snail) cDNA library1,4. ω-CTx-MVIID blocks Ca2+ current activated in whole-cell patch-clamp recordings from central neurons of goldfish2. ω-conotoxin MVIID exhibits greater discrimination against the N-channel subtype than any other ω-conotoxin variant to date. ω-conotoxin MVIID has effects on “P-like” subtypes of Ca2+ channels. Furthermore, ω-CTx-MVIID inhibits synaptosomal voltage-sensitive Ca2+ uptake3.
