- Alomone Labs ω-Conotoxin SO3 inhibits CaV2.2 channels heterologously expressed in Xenopus oocytes.A. Time course of ω-Conotoxin SO3 (#STC-470) blocking action on CaV2.2 currents (α1B + α2δ1 + β1). Maximum current amplitude was plotted as a function of time. Membrane potential was held at -100 mV and cells were stimulated in the presence of 2 mM Ba2+ by a 100 ms voltage ramp to +60 mV. 50 nM ω-Conotoxin SO3 were perfused as indicated by the bar (green) during 150 s. B. Superimposed examples of CaV2.2 channel current in the absence (control) and presence (green) of 50 nM ω-Conotoxin SO3 (taken from the experiment in A).

Scientific background ω-conotoxin SO-3 is a peptide toxin originally isolated from the venom of Conus striatus. It is a selective blocker of N-type CaV channels. SO-3 inhibits INa and IK with IC50 values of 0.49 and 1.6 mM respectively. ω-conotoxin SO-3 does not show any inhibiting effects on L-type, P/Q-type and R-type currents at 3 µM concentration and has no effect on NaV currents, delayed rectifier K+ currents and transient outward K+ currents. ω-conotoxin SO-3 displays an analgesic potency similar to ω-conotoxin MVIIA in a range of acute and chronic pain models in rodents, but has less adverse effects compared with identical dosages of ω-conotoxin MVIIA injected intrathecally1.
