Activity 1-Norokadaone is an inactive analogue of okadaic acid (a PP1 and PP2A inhibitor) even at high concentrations1. It may be used as a negative control vis-à-vis okadaic acid.

Our bioassay
- Alomone Labs 1-Norokadaone serves as a negative control for Okadaic Acid in modulating the phophatase activity of PP-2A.Cells were grown to 70% confluence and serum starved for 18 h. For in vivo phosphatase activity, the growth medium was replaced with HENK’s and the cells were preincubated in the presence or absence of 100 nM Okadaic Acid (ammonium salt) (#O-800), (or Okadaic Acid (sodium salt) (#O-900)) and 100 nM 1-Norokadaone (#N-750) followed by a 20 min incubation with or without 20 µM NEM. The cells were then stimulated with 5 ng/ml PDGF for 30 min. At the end of the incubation, cell proteins were resolved by SDS-PAGE and probed with anti-phospho-Akt antibodies.

Scientific background 1-Norokadaone is structural analogue of Okadaic acid. This compound is a less active homologue than the Okadaic Acid and Okadaol as inhibitors of Protein Phosphatase 1 and Protein Phosphatase 2A, and is thus widely used as negative control for Okadaic Acid1-3.
