Overview
- Peptide (C)EISPGKINNVEQND, corresponding to amino acid residues 158 - 171 of mouse SLC9A7 (Accession Q96T83). Extracellular, 2nd loop.
- Western blot analysis of rat brain membranes (lanes 1 and 3) and mouse brain membranes (lanes 2 and 4):1-2. Anti-Na+/H+ Exchanger 7 (NHE7) (extracellular) Antibody (#ANX-007), (1:200).
3-4. Anti-Na+/H+ Exchanger 7 (NHE7) (extracellular) Antibody, preincubated with Na+/H+ Exchanger 7 (NHE7) (extracellular) Blocking Peptide (BLP-NX007). - Western blot analysis of human U-87 MG glioma cell line lysate:1. Anti-Na+/H+ Exchanger 7 (NHE7) (extracellular) Antibody (#ANX-007), (1:200).
2. Anti-Na+/H+ Exchanger 7 (NHE7) (extracellular) Antibody, preincubated with Na+/H+ Exchanger 7 (NHE7) (extracellular) Blocking Peptide (BLP-NX007).
- Expression of NHE7 in mouse hippocampus.Immunohistochemical staining of perfusion-fixed frozen mouse brain sections with Anti-Na+/H+ Exchanger 7 (NHE7) (extracellular) Antibody (#ANX-007), (1:200), followed by goat anti-rabbit-AlexaFluor-488. A. Staining in the mouse hippocampal CA3 region, showed immunoreactivity of NHE7 (green) in the CA3 pyramidal layer (arrows). B. Pre-incubation of the antibody with Na+/H+ Exchanger 7 (NHE7) (extracellular) Blocking Peptide (BLP-NX007), suppressed staining. Cell nuclei are stained with DAPI (blue).
- Cell surface detection of NHE7 by indirect flow cytometry in live intact human U-87 MG glioma line:___ Cells.

Na+/H+ exchangers (NHEs) also termed solute carrier family 9 (SLC9), are proteins that function as ion transporters. NHEs are present across all kingdoms of life from prokaryotes to eukaryotes and are found to be crucial in preventing intracellular acidification. NHEs are divided into 3 groups SLC9A (NHE1-9), SLC9B (NHA1 and NHA2) and SLC9C according to their cellular location and their function. NHEs regulates cell volume and pH of the cytosol and organelles1.
NHE7 is an NHE that is localized predominantly to the trans-Golgi network (TGN) and post-Golgi vesicles. NHE7 differ from other NHEs by its ability to transport both Na+ and K+ via a H+ gradient-dependent manner2. NHE7 mRNA is expressed in wide range of cells but is especially abundant in the brain, skeletal muscle and various secretory tissues3.
The predicted membrane topology of NHE7 consist of 12 transmembrane domains (TMDs) and a single N-glycosylation site at position 145. Similarly to the other SLC9A isoforms NHE7 can be assembled as a homodimer. It was recently found that a missense variant in NHE7 associates with multigenerational nonsyndromic intellectual disability (ID)3.

Anti-Na+/H+ Exchanger 7 (NHE7) (extracellular) Antibody (#ANX-007) is a highly specific antibody directed against an extracellular epitope of the mouse protein. The antibody can be used in western blot and immunohistochemistry applications. It has been designed to recognize NHE7 from rat, mouse and human samples.
