Solute carrier family 28 member 3 (SLC28A3, CNT3) is a sodium-dependent, concentrative pyrimidine- and purine-selective transporter. SLC28A3 plays an important role in mediating the cellular entry of a broad array of physiological nucleosides and synthetic anticancer nucleoside analog drugs, in a process that is coupled to the sodium cellular gradient1.
CNT3 consists of 13 putative trans-membrane domains and it forms a homo-trimer which is important for its activity as a transporter2,3.
SLC28 members are crucial determinants in the response to a variety of anticancer and antiviral nucleoside analogs, as they modulate the entry of these analogs into target tissues. Within this large group of solute carriers, SLC28A is a sub-group which consists of SLC28A1, SLC28A2, and SLC28A3. Of the three members of this sub-family, SLC28A3 has the broadest selectivity and the widest expression profile4.
