SLC35G1 (Solute carrier family 35 member G1) is a 10 transmembrane-spanning protein that interacts with STIM1 and is involved in calcium level regulation in the ER, plasma membrane and mitochondria1.
Stromal interaction molecule 1 (STIM1) translocates within the ER membrane upon calcium depletion and interacts with Orai1 on the plasma membrane. STIM1 gates Orai1, allowing calcium to enter the cytoplasm, where it repletes the ER store via Calcium-ATPase pumps.
SLC35G1 is located on the plasma membrane and ER. Upon calcium depletion, SLC35G1 binds STIM1 and moves it within the ER to localize it near the cell membrane. hence its role in regulating cellular calcium levels1,2.
Due to its involvement in the STIM1-Orai1 axis, SLC35G1 mutations may have a role in immunodeficiency, muscular hypotonia, Stormorken syndrome and York platelet syndrome and many other calcium flux related syndromes3.
