Our bioassay
- Alomone Labs Hemitoxin inhibits KV1.2 channel currents heterologously expressed in Xenopus oocytes.Currents were elicited by application of a 100 msec voltage ramp from a holding potential of -80 mV to 60 mV, delivered every 10 seconds. A. Time course of channel activity (current amplitude at -20 mV), before and during application of 500 and 100 nM Hemitoxin (#STH-500) as indicated by the bars. B. Example of superimposed current traces before and during application of 500 and 100 nM Hemitoxin (taken from the experiment in A).
Scientific background Hemitoxin is a 35 amino acid peptidyl toxin isolated from the Hemiscorpius lepturus scorpion venom and belongs to the αKTx6 family1. It reversibly blocks KV1.1, KV1.2 and KV1.3 voltage-gated K+ channels with an IC50 of 13 nM, 16 nM and 2 nM, respectively1. It shares 80% sequence similarity with Maurotoxin, and has a unique pairing of its four disulfide bridges1.
Target KV1.1, KV1.2, KV1.3 K+ channels
