Our bioassay
- Alomone Labs Lq2 inhibits Kir1.1 (ROMK1) channels heterologously expressed in Xenopus oocytes.A. ROMK1 channel current (in 5 mM K+) elicited by 200 ms depolarization from a holding potential of -100 mV to +20 mV, before and during application of Lq2 (#RTL-550) at the indicated concentrations. B. Mean ±S.D. dose response for Lq2 inhibition of ROMK1 channels (n = 4-6 oocytes for each time point). The smooth line is a fit giving a Ki of 5 nM. C. Mean ± S.D. Scientific background
Lq2 was originally isolated from Leiurus quinquestriatus hebraeus scorpion venom.1
Lq2 blocks Kir1.1 K+ inward rectifier channels (Ki of 410 nM).2
In the planar lipid bilayer, it also blocked single KCa channels from rat muscle with intrinsic KD of 43 nM,1 by introducing brief closures. Lq2 action is on the extracellular side of the pore.
