Activity PMA is a commonly used phorbol ester and activates PKC in nM concentrations1. It has also been shown to be an extremely potent tumor promoter2-4.

- Alomone Labs PMA activates PKC in NIH-3T3 cells.Cells were stimulated with 0.6 µM PMA (#P-800) for 20 min. Cell proteins were resolved by SDS-PAGE and probed with anti-phospho (ser)- MARCKS (myristoylated alanin-rich protein kinase C substrate).
Scientific background Phorbol 12-myristate 13-acetate (PMA) is the most commonly-used phorbol ester, primary found as extremely potent tumor promoter.1-3 PMA is a reversible activator of Protein Kinase C (PKC)4 in vivo and in vitro, even at nM concentrations (cells are usually treated with PMA at 200 nM to induce signaling). PMA promotes the expression of inducible NOS in cultured hepatocytes, activates Ca2+-ATPase pump and potentates forskolin-induced cAMP formation. In some cases PMA inhibits apoptosis induced by the Fas antigen, but induces apoptosis in HL-60 promyelocytic leukemia cells.5-8
