Our bioassay
Alomone Labs WS 12 activates TRPM8 channels expressed in HEK-293 cells.Dose response curve for the activation of human TRPM8 by rising concentrations of WS 12 (#W-105), showing activity at low nanomolar range. Cells were loaded with Calcium-5 dye. Changes in intracellular Ca2+ were detected as changes in 485 nm/525 nm fluorescence following agonist application.
Scientific background
The carboxamide WS 12 is a synthetic selective TRPM8 channel agonist1.
TRPM8 is a Ca2+-permeable channel, a member of the ‘long' or melastatin (M) group of TRP proteins and is activated by temperatures or by cooling compounds such as icilin, menthol and eucalyptol. The TRPM8 gene is expressed and is up-regulated in prostate cancer and other malignancies. It is specifically expressed in temperature-sensing trigeminal and dorsal root ganglion neurons1-3.
